干细胞诱导分化为视网膜色素上皮细胞的研究进展

摘要:

近年来干细胞替代治疗在眼科领域的应用受到越来越多的关注,胚胎干细胞、诱导多能干细胞及成体干细胞作为细胞替代治疗的种子细胞, 能被定向诱导分化为视网膜色素上皮细胞,为视网膜退行性疾病的细胞替代治疗提供丰富的细胞来源。最近已有大量研究者能通过干细胞自我分化、共培养、加入特定因子、细胞再编程等获得视网膜色素上皮细胞。此外体内移植干细胞来源的视网膜色素上皮细胞已被证明能改善动物视功能,更重要的是美国政府已批准胚胎干细胞来源的视网膜色素上皮细胞用于临床试验。本文将对胚胎干细胞、诱导多能干细胞及成体干细胞诱导分化成视网膜色素上皮细胞的方法、诱导后结果及临床应用等进行综述。(国际眼科纵览, 2016,  40:  295-300)

Abstract:

In recent years, more and more attention is paid to stem cell-replacement therapy in the field of ophthamology. Stem cells including embryonic stem cells
(ESCs), induced pluripotent stem cells (iPSCs), and adult stem cells, possessing the ability of differentiation towards retinal pigment epithelium (RPE), can provide an unlimited source of replacement retinal pigment epithelium cells for retinal degenerative disease. Recently, a large number of researchers have demonstrated that RPE cells could be obtained from stem cells by spontaneous differentiation, coculturing, defined factors or cell reprogramming, respectively. In addition, in vivo studies also showed that the restoration of visual function could be obtained by transplanting functional RPE cells into the subretinal space of animal model. What is more, clinical trials approved by the USA have shown promising prospects in RPE transplantation. In this review, we will discuss the various protocols for generating RPE cells from ESCs, iPSCs, and MSCs and their research progress. (Int Rev Ophthalmol, 2016, 40: 295-300)
