Metabonomics and its application in ophthalmology

摘要: 代谢组学是定性和定量地检测机体受到病理生理刺激或者发生基因修饰后所有低分子量代谢产物的一门新兴学科。通过对内源性代谢产物的变化规律进行分析,不仅可以发现用于疾病早期诊断的新的生物标记物,还可以探索疾病的发病机制。代谢组学常用的研究技术包括核磁共振技术和质谱法,常见的统计分析方法包括非监督分析方法和监督法。本文回顾了代谢组学在临床疾病的诊断及药物毒理学研究领域的应用,并着重对其在眼科相关疾病如眼外伤、葡萄膜炎、白内障、高眼压症及视神经病变在早期诊断、鉴别诊断、发病机制和药物疗效等方面的研究进展做一综述。

Abstract: Metabonomics is a new discipline defined as the qualitative and quantitative measurement of low-molecular-weight metabolites in organisms subject to pathophysiological stimuli or genetic modification. This method enables us to have a new perspective on metabolic variation. It provides the possibility of exploring the biomarkers of diseases for early diagnosis and pathogenesis of diseases. The research techniques used in metabonomics include nuclear magnetic resonance (NMR) technique and mass spectrometry (MS). The unsupervised method and supervised method are useful statistical techniques for the analysis of metabonomic data. In this article, the application of metabolomics in clinical diagnosis and drug toxicology are reviewed briefly with an emphasis laid on its progress in ophthalmology. We focused on the research progress of metabolomics in early diagnosis, differential diagnosis, pathogenesis and drug efficacy of particularly relevance to eye diseases such as ocular trauma, uveitis, cataracts, ocular hypertension and optic neuropathy.
